چکیده

We describe the development of a novel protein expression system based on the industrial fermentation organism Ralstonia eutropha (formerly known as Alcaligenes eutrophus) NCIMB 40124. This new system overcomes some of the shortcomings of traditional Escherichia coli-based protein expression systems, particularly the propensity of such systems to form inclusion bodies during high-level expression. Using a proteomics approach, we identified promoters that can be induced by simple process parameters or medium compositions in high-density cell culture or shake flasks, respectively. By combining newly developed molecular biological tools with a high-cell-density fermentation process, we were able to produce high levels (>1 g/liter) of soluble, active organophosphohydrolase, a model enzyme prone to inclusion body formation in E. coli.

High level recombinant protein expression in Ralstonia eutropha using T7 RNA polymerase based amplification.

We report further development of a novel recombinant protein expression system based on the Gram-negative bacterium, Ralstonia eutropha. In this study, we were able to express soluble, active, organophosphohydrolase (OPH), a protein that is prone to inclusion body formation in Escherichia coli, at titers greater than 10 g/L in high cell density fermentation. This represents a titer that is appr...

Influence of Matric Potential on Survival and Activity of Genetically Engineered Ralstonia eutropha H850Lr

Although the application of biodegradative genetically engineered micro organisms (GEMs) for bioremediation is very promising, the risks of their release should be assessed before their introduction into the environment. Lux-marked Ralstonia eutropha H850Lr (formerly Alcaligenes eutrophus H850Lr) was introduced into sterile and non-sterile soil microcosms at matric potentials ?2.11, ?30, ?750, ...

Autotrophic production of stable-isotope-labeled arginine in Ralstonia eutropha strain H16.

With the aim of improving industrial-scale production of stable-isotope (SI)-labeled arginine, we have developed a system for the heterologous production of the arginine-containing polymer cyanophycin in recombinant strains of Ralstonia eutropha under lithoautotrophic growth conditions. We constructed an expression plasmid based on the cyanophycin synthetase gene (cphA) of Synechocystis sp. str...

Aerobic degradation of methylene blue from colored effluents by Ralstonia eutropha

The present paper has examined the degrading ability of phenol-oxidizing bacterium, Ralstonia eutropha, for biological removal of methylene blue (MB) from aqueous solutions under aerobic conditions. Results show that MB has been extensively eliminated as a co-metabolism in the presence of supplementary carbon (glucose) and nitrogen (yeast extract and peptone) sources and the experimental observ...

Integrated recombinant protein expression and purification platform based on Ralstonia eutropha.

Protein purification of recombinant proteins constitutes a significant cost of biomanufacturing and various efforts have been directed at developing more efficient purification methods. We describe a protein purification scheme wherein Ralstonia eutropha is used to produce its own "affinity matrix," thereby eliminating the need for external chromatographic purification steps. This approach is b...
